A new superweapon in the fight against cancer

    especially the ones that come back
    over and over again.

    The highly invasive
    and drug-resistant ones,
  • Not Synced
    the ones that defy medical treatment,
  • Not Synced
    even when we throw our best drugs at them.
  • Not Synced
    Engineering at the molecular level,
  • Not Synced
    working at the smallest of scales,
  • Not Synced
    can provide exciting new ways
  • Not Synced
    to find the most aggressive
    forms of cancer.
  • Not Synced
    Cancer is a very clever disease.
  • Not Synced
    There are some forms of cancer,
  • Not Synced
    which, fortunately, we've learned
    how to address relatively well
  • Not Synced
    with known and established drugs
    and surgery.
  • Not Synced
    But, there's some forms of cancer
    that don't respond
  • Not Synced
    to these approaches
  • Not Synced
    and the tumor survives
    or comes back,
  • Not Synced
    even after an onslaught of drugs.
  • Not Synced
    We can think of these
    very aggresive forms of cancer
  • Not Synced
    as kind of super villains in a comic book.
  • Not Synced
    They're clever, they're adaptable,
  • Not Synced
    and they're very good at staying alive.
  • Not Synced
    And, like most super villains
    these days,
  • Not Synced
    their super powers come from
    a genetic mutation.
  • Not Synced
    The genes that are modified
    inside these tumor cells
  • Not Synced
    can enable and encode for new
    and unimagined modes of survival,
  • Not Synced
    allowing the cancer cell
    to live through
  • Not Synced
    even our best chemotherapy treatments.
  • Not Synced
    One example is a trick
    in which a gene allows
  • Not Synced
    a cell, even as the drug
    approaches the cell,
  • Not Synced
    to push the drug out before the drug
    can have any effect.
  • Not Synced
    Imagine the cell effectively
    spits out the drug.
  • Not Synced
    This is just one example
    of the many genetic tricks
  • Not Synced
    in the bad of our super villain, cancer.
  • Not Synced
    All due to mutant genes.
  • Not Synced
    So, we have a super villain
    with incredible super powers
  • Not Synced
    and we need a new and
    powerful mode of attack.
  • Not Synced
    Actually, we can turn off a gene,
  • Not Synced
    the key is a set of molecules
    called siRNA.
  • Not Synced
    siRNA are short sequences
    of genetic code
  • Not Synced
    that guide a cell to block
    a certain gene.
  • Not Synced
    Each siRNA molecule
    can turn off a specific gene
  • Not Synced
    inside the cell.
  • Not Synced
    For many years since its discovery,
  • Not Synced
    scientists have been very excited
  • Not Synced
    about how we can apply
    these gene blockers in medicine.
  • Not Synced
    But, there is a problem.
  • Not Synced
    siRNA works well inside the cell.
  • Not Synced
    But if it gets exposed to the enzymes
    that reside
  • Not Synced
    in our bloodstream and our tissues,
  • Not Synced
    it degrades within seconds.
  • Not Synced
    It has to be packaged, protected
    through its journey through the body
  • Not Synced
    on its way to its final target
    inside the cancer cell.
  • Not Synced
    So, here's our strategy:
  • Not Synced
    first, we'll dose the cancer cell
    with siRNA, the gene blocker,
  • Not Synced
    and silence those viral genes,
  • Not Synced
    and they'll we'll whap (?) it
    with a chemo drug.
  • Not Synced
    But how do we carry that out?
  • Not Synced
    Using molecular engineering,
  • Not Synced
    we can actually design
    a super weapon
  • Not Synced
    that can travel through the blood stream.
  • Not Synced
    It has to be tiny enough
    that it can get through the blood stream,
  • Not Synced
    it's got to be small enough
    to penetrate the tumor tissue,
  • Not Synced
    and it's got to be tiny enough
    to be taken up
  • Not Synced
    inside the cancer cell.
  • Not Synced
    To do this job well, it has to be
    about one 100th the size
  • Not Synced
    of a human hair.
  • Not Synced
    Let's take a closer look
    at how we can build this nanoparticle.
  • Not Synced
    First, let's start with
    the nanoparticle core.
  • Not Synced
    It's a tiny capsule that contains
    the chemotherapy drug.
  • Not Synced
    This is the poison that will
    actually end the tumor cell's life.
  • Not Synced
    Around this core, we'll wrap
    a very thin,
  • Not Synced
    nanometer-think blanket
    of siRNA.
  • Not Synced
    This is our gene blocker.
  • Not Synced
    Because siRNA is strongly negatively charged,
  • Not Synced
    we can protect it with a nice
    protect layer
  • Not Synced
    of postively charged polymer.
  • Not Synced
    The two oppositely charged molecules
    stick together throough charge attracttion,
  • Not Synced
    and that provides us with a protective
    layer
  • Not Synced
    that prevents the sIRNA from
    degrading the blood stream.
  • Not Synced
    We're almost done.
  • Not Synced
    (Laughter)
  • Not Synced
    But there is one more big obstacle
    we have to think about.
  • Not Synced
    In fact, i tmay be the biggest
    obstacle of all.
  • Not Synced
    How do we deploy this super weapon?
  • Not Synced
    I mean, every good weapon
    needs to be targeted,
  • Not Synced
    we need to target this super weapon
    to the super villain cells
  • Not Synced
    that we find in the tumor.
  • Not Synced
    But, our bodies have a natural
    immune defense system.
  • Not Synced
    Cells that reside in teh blood stream
  • Not Synced
    and pick out things that don't belong
  • Not Synced
    so that it can destroy or elinate them.
  • Not Synced
    And guess that, our nanoparticle
    is considered a foreign object.
  • Not Synced
    We have to sneak our nanoparticle
  • Not Synced
    past the tumor dfense system,
  • Not Synced
    we have to get it past this mechanism
    of getting rid of the foreign object
  • Not Synced
    bu disguising it.
  • Not Synced
    So we add one more negatively charged layer
  • Not Synced
    around this nanoparticle,
  • Not Synced
    which serves two purposes.
  • Not Synced
    First, this outer layer is one of
    the naturally charged,
  • Not Synced
    highly hydrated polysaccarides
    that resides in our bodies.
  • Not Synced
    It creates a cloud of water molecules
    around the nanoparticle
  • Not Synced
    that gives us an invisibility cloaking affect.
  • Not Synced
    This invisibility cloak allows
    the nanoparticle
  • Not Synced
    to travel through the bloodstream
    long and far enough
  • Not Synced
    to reach the tumor without getting
    eliminated by the body.
  • Not Synced
    Second, this layer contains molecules
    which bind specifically
  • Not Synced
    to our tumor's cell.
  • Not Synced
    Once bound, the cancer cell takes up
    the nanoparticle
  • Not Synced
    and now we have our nanoparticle
    inside the cancer cell
  • Not Synced
    and ready to deploy.
  • Not Synced
    Alright, I feel the same way,
    let's go.

    The siRNA IS DEPLOYED FIRST.

    It acts for hours,
  • Not Synced
    giving enough time to silence
    and block those survival genes.
  • Not Synced
    We have now disabled those
    genetic superpowers.

    What remains is a cancer cell
    with no special defenses.

    Then, the chemotherapy drug
    comes out of the core
  • Not Synced
    and destroys the tumor cell
    cleanly and efficiently.

    With sufficient gene blockers,
  • Not Synced
    we can address many different kinds
    of mutations.

    Allowing the chance to sweep out tumors,
    without leaving behind any bad guys.

    So, how does our strategy work?

    We've tested these nano-strucutre particles
    in animals
  • Not Synced
    using a highly aggresive form
    of triple negative breast cancer.
  • Not Synced
    This triple negative breast cancer
    exhibits the gene
  • Not Synced
    that spits out cancer drugs
    as soon as its delivered.
  • Not Synced
    Usually, ?, let's call it ?
  • Not Synced
    is the cancer drug that is
    the first line of treatment
  • Not Synced
    for breast cancer.
  • Not Synced
    So, we first treated our animals
    with dox core, dox only.
  • Not Synced
    The tumor slowed their rate of growth,
  • Not Synced
    but they still grew rapidly,
  • Not Synced
    doubling in size over a period of two weeks.
  • Not Synced
    Then, we tried our combination super weapon.
  • Not Synced
    And now layer a particle
    with siRNA against the chemo pump,
  • Not Synced
    plus, we have the doxs in the core.
  • Not Synced
    And look, we found that not only
    did the tumors stop growing,
  • Not Synced
    they actually decerased in size
  • Not Synced
    and were elimintaed
    in some cases.
  • Not Synced
    The tumors were actually
    regressin.
  • Not Synced

    What's great about this approach
    is that it can be personalized,

    we can add many different layers
    of siRNA
  • Not Synced
    to address different mutations
    and tumor defense mechanisms
  • Not Synced
    and we can put different drugs
    into the nanoparticle core.

    As doctors learn how to test patients
    and understand
  • Not Synced
    certain tumor genetic types,
  • Not Synced
    they can help us determine
    which patients
  • Not Synced
    can benefit from this strategy
  • Not Synced
    and which gene blockers we can use.

    Ovarian cancer strikes
    a special chord with me.

    It is a very aggresive cancer,
  • Not Synced
    in part because it's discovred
    at very late stages
  • Not Synced
    when its highly advanced
  • Not Synced
    and there are a number
    of genetic mutations.

    After the first round of chemotherapy,
  • Not Synced
    this cancer comes back
    for 75 percent of patients.
  • Not Synced
    And it usually comes back
    in a drug resistant form.

    High-grade ovarian cancer
    is one of
  • Not Synced
    the biggest super villains out there.
  • Not Synced
    And we're now directlng
    this super weapon
  • Not Synced
    towards its defeat.
  • Not Synced
    As a researcher,
  • Not Synced
    I usually don't get to work with patients,
  • Not Synced
    but I recently met a mother
  • Not Synced
    who is an ovarian cancer survivor,
    Mimi and her daughter pAIGE.
  • Not Synced
    I was deeply inspired
  • Not Synced
    by the optimism and strength
  • Not Synced
    that both mother and daughter displayed.
  • Not Synced
    And by their story of courage and support.
  • Not Synced
    At this event, we spoke about
    the different technologies
  • Not Synced
    directed at cancer.
  • Not Synced
    And Mimi was in tears
    as she explained how
  • Not Synced
    learning about these efforts
  • Not Synced
    gives her hope for future generations,
  • Not Synced
    including her own daughter.
  • Not Synced
    This really touched me.
  • Not Synced
    It's not just about building
    really elegant science,
  • Not Synced
    it's about changing people's lives.
  • Not Synced
    It's about understanding the power
    of engineering
  • Not Synced
    on the scale of molecules.
  • Not Synced
    I know that as students like Paige
  • Not Synced
    move forward in their careers,
  • Not Synced
    they'll open new possibilites
  • Not Synced
    in addressing some of the big
    health problems in the world.

    Including, ovarian cancer,
    neurological disorders,
  • Not Synced
    and infectious disease.
  • Not Synced
    Just as chemical engineering
    has found a way
  • Not Synced
    to open doors for me.

    Has provided a way of engineering
    on the tiniest scale

    that of molecules
  • Not Synced
    to heal on the human scale.
