    an old technology
    that of course, you all know and love,

    and that's called drugs.

    But I'll come to that in a minute.

    I'm going to start with the brain
  • 0:40 - 0:43
    because it's the most complex
    and evolved element
  • 0:43 - 0:45
    in the whole universe.
  • 0:45 - 0:49
    It's what got you here today,
    and hopefully, will get you home tonight.
  • 0:49 - 0:52
    A single mouse brain has
    more computing power
  • 0:52 - 0:54
    than all the computers on Earth today.
  • 0:54 - 0:59
    Your brains are at least
    a million times more powerful.
  • 1:00 - 1:02
    But unfortunately, it can go wrong.
  • 1:02 - 1:04
    Over the last few years,
  • 1:04 - 1:10
    we've discovered the scale of problems
    that brain disorders produce.
  • 1:11 - 1:17
    The sum total of illness and cost
    to society from brain disorders
  • 1:17 - 1:20
    is greater than that from cancer,
  • 1:20 - 1:23
    cardio-vascular disease,
    and diabetes put together.
  • 1:23 - 1:24
    You see on the graph there
  • 1:24 - 1:29
    it's the equivalent each year
    to nearly 800 billion euros.
  • 1:29 - 1:32
    It is if we're paying off
    the Greek debt every year
  • 1:32 - 1:36
    in the burden of illness
    produced by brain disorders.
  • 1:36 - 1:42
    We know that investment in these disorders
    is not matching the enormous burden.
  • 1:42 - 1:47
    Here, you can see
    on the left hand graph, the red circle;
  • 1:47 - 1:52
    'brain disorders' are way outside
    the predicted line of investment.
  • 1:52 - 1:54
    They're the largest disability,
  • 1:54 - 1:57
    and the investment is
    disproportionately low.
  • 1:57 - 2:00
    On the right-hand side, you can see
    one of the reasons for this.
  • 2:00 - 2:03
    You can see the attrition rate for drugs
  • 2:03 - 2:07
    that go through discovery
    into development.
  • 2:07 - 2:09
    Look at the second cylinder there.
  • 2:09 - 2:15
    You can see that from
    200 Alzheimer's drugs in development,
  • 2:15 - 2:18
    only one reaches the clinic.
  • 2:18 - 2:21
    The brain is
    a very difficult organ to treat.
  • 2:23 - 2:24
    Why does the brain go wrong?
  • 2:24 - 2:27
    It goes wrong
    because of external influences:
  • 2:27 - 2:29
    malnutrition, still a big problem;
  • 2:29 - 2:33
    parental and other abuse
    - psychological and physical -;
  • 2:33 - 2:36
    toxins - particularly alcohol.
  • 2:36 - 2:40
    These are images of my own research
    showing a normal brain at the top
  • 2:40 - 2:43
    and a brain severely damaged
    by alcohol misuse lower down.
  • 2:43 - 2:47
    Infections such as meningitis,
    encephalitis are still common,
  • 2:47 - 2:50
    and trauma is a massive problem
  • 2:50 - 2:53
    in terms of leading
    to long-term brain damage
  • 2:53 - 2:55
    particularly in young men.
  • 2:55 - 2:58
    And then, there are internal aspects
    of the brain development
  • 2:58 - 3:01
    that can go wrong:
    related conditions like autism.
  • 3:01 - 3:04
    You can have acquired
    abnormalities like epilepsy
  • 3:04 - 3:07
    and there are age-related changes
    such as dementia.
  • 3:10 - 3:14
    But the real focus of my talk today
    is how the brain limits itself
  • 3:14 - 3:19
    and how we can perhaps expand its capacity
    or take away the limit it puts on itself.
  • 3:19 - 3:22
    Your brain is most flexible
    when you're a baby.
  • 3:22 - 3:24
    Some people would argue
  • 3:24 - 3:28
    that the whole process of education
    is about taking away flexibility
  • 3:28 - 3:33
    and forcing every one of you
    to think and behave in the same way.
  • 3:33 - 3:35
    It's about getting conformity of process
  • 3:35 - 3:37
    which of course is useful
  • 3:37 - 3:40
    if you're trying to speak a language
    the same way as other people,
  • 3:40 - 3:42
    but may not be useful
  • 3:42 - 3:46
    if it limits how you can deal with
    other things such as problems.
  • 3:46 - 3:50
    And also, the constraining of the brain
    in itself can lead to problems;
  • 3:50 - 3:52
    if there is not enough of it
    in the right place,
  • 3:52 - 3:56
    you get disorders
    such as ADHD and schizophrenia.
  • 3:56 - 3:58
    If you get excessive constraints,
  • 3:58 - 4:01
    you can end up with disorders
    like OCD and addiction.
  • 4:01 - 4:06
    And also the resilience
    in the brain can be impaired,
  • 4:06 - 4:10
    and that will lead to disorders
    such as anxiety and depression.
  • 4:10 - 4:16
    The core of my talk really is showing you
    how we can now understand
  • 4:16 - 4:20
    the limitations that the brain
    constrains the mind with
  • 4:20 - 4:22
    through using drugs.
  • 4:22 - 4:25
    This research really goes back
    to the 1950s
  • 4:25 - 4:29
    and the personal experience
    of this man, Aldous Huxley,
  • 4:29 - 4:32
    who used peyote and used
    LSD, psychedelic drugs,
  • 4:32 - 4:34
    to understand his mind.
  • 4:34 - 4:37
    He wrote about it in the book
    "The Doors of Perception,"
  • 4:37 - 4:40
    and he used this quote from William Blake
  • 4:40 - 4:44
    to explain how these drugs
    changed his mind.
  • 4:44 - 4:47
    He said, "If the doors
    of perception were cleansed,
  • 4:47 - 4:50
    everything would appear
    to man as it is, Infinite.
  • 4:50 - 4:52
    For man has closed himself up
  • 4:52 - 4:57
    till he sees all things through
    narrow chinks of his cavern."
  • 4:57 - 5:01
    And Huxley realized
    that what psychedelics do
  • 5:01 - 5:04
    is take away this phenomenon
    that he inferred
  • 5:04 - 5:09
    which is that "the brain is
    an instrument for focusing the mind."
  • 5:09 - 5:12
    Modern neuroscience
    has shown they were right.
  • 5:12 - 5:15
    Because, what we now know
  • 5:15 - 5:19
    is that the brain creates
    what the mind thinks it's doing.
  • 5:19 - 5:21
    Here is an example of vision.
  • 5:21 - 5:24
    You might be looking out
    at a glorious sunset,
  • 5:24 - 5:28
    but in reality, the light rays
    go into your retina
  • 5:28 - 5:31
    and are transformed
    into a series of electrical impulses
  • 5:31 - 5:34
    which pass into parts in your brain.
  • 5:34 - 5:37
    And those parts of the brain
    reconstruct an image
  • 5:37 - 5:40
    that they think you're seeing.
  • 5:41 - 5:44
    And that image, in the words of Blake,
  • 5:44 - 5:47
    is seeing through the constraints,
    the chinks of the cavern
  • 5:47 - 5:49
    that your brain puts on it.
  • 5:50 - 5:55
    And if you have mental illness
    - for instance, depression or addiction -
  • 5:55 - 5:59
    then what you see is also
    constrained by your brain.
  • 5:59 - 6:02
    So depressed people don't see
    even the brightness of the sky.
  • 6:02 - 6:04
    They see a dull grey.
  • 6:04 - 6:07
    And of course, people with addiction
  • 6:07 - 6:10
    when they see through
    the chinks of their cavern,
  • 6:10 - 6:13
    they simply see the drugs
    that they're addicted to.
  • 6:15 - 6:19
    Psychedelic drugs
    take away that limitation.
  • 6:19 - 6:23
    They allow the mind to work
    in a much more flexible way.
  • 6:23 - 6:26
    This is our research using psilocybin
    in magic mushroom juice.
  • 6:26 - 6:30
    Those two images contain
    the same number of connections.
  • 6:30 - 6:32
    But on the left-hand image, under placebo,
  • 6:32 - 6:36
    you see that most of the connections
    are around the edge.
  • 6:36 - 6:40
    The brain talks to itself
    in regional ways.
  • 6:40 - 6:41
    But under psilocybin,
  • 6:41 - 6:45
    there's a massive cross-talk,
    much more integration;
  • 6:45 - 6:48
    parts of the brain which haven't talk
    to each other since you were children
  • 6:48 - 6:50
    are able to engage.
  • 6:50 - 6:52
    And that's how people can get new insights
  • 6:52 - 6:57
    and also, potentially overcome damage
    of dysfunction of the brain.
  • 6:57 - 7:01
    Here is another study using LSD
    showing essentially the same thing.
  • 7:01 - 7:03
    On the left-hand side,
  • 7:03 - 7:08
    you see the visual cortex is
    normally very local in how it works.
  • 7:08 - 7:11
    But under LSD,
    when people have their eyes closed,
  • 7:11 - 7:16
    they can see enormously vivid,
    interesting sets of images.
  • 7:16 - 7:19
    And that's because the brain is
    much more interconnected under LSD
  • 7:19 - 7:21
    than normal.
  • 7:21 - 7:24
    Here you see that the visual cortex
  • 7:24 - 7:27
    connects to most of the rest
    of the brain in that state.
  • 7:28 - 7:34
    We've been able to utilize this liberation
    of processing of the brain,
  • 7:34 - 7:35
    produced by psychedelics,
  • 7:35 - 7:37
    to treat people with depression.
  • 7:37 - 7:39
    Here is a study published last year
  • 7:39 - 7:42
    where we took people with depression
  • 7:42 - 7:44
    who failed two previous
    anti-depressive treatments,
  • 7:44 - 7:47
    and also had failed psychotherapy.
  • 7:47 - 7:49
    They were given
    a single dose of psilocybin,
  • 7:49 - 7:52
    and you can see there, that a week later,
  • 7:52 - 7:54
    all of them had recovered to some extend,
  • 7:54 - 7:57
    and half of them were now
    in a state of remission.
  • 7:57 - 7:59
    They were in the yellow bar there,
  • 7:59 - 8:02
    which shows that their depression
    has actually gone away.
  • 8:02 - 8:04
    And that's not the first evidence
  • 8:04 - 8:08
    that psychedelics have therapy
    or have therapeutic uses.
  • 8:08 - 8:10
    We also have evidence
    from around the world
  • 8:10 - 8:14
    that psilocybin can be useful in helping
    people deal with alcohol dependence,
  • 8:14 - 8:18
    with smoking dependence,
    with obsessive compulsive disorder.
  • 8:18 - 8:20
    And most recently,
  • 8:20 - 8:23
    two major studies showing
    it can help people come to terms
  • 8:23 - 8:25
    with the anxiety and the depression
  • 8:25 - 8:30
    which almost always accompany
    a diagnosis of a terminal illness.
  • 8:30 - 8:33
    So, these drugs can have
    potentially enormous opportunities
  • 8:33 - 8:36
    for helping people deal
    with mental distress.
  • 8:37 - 8:40
    It's not just psychedelics
    that can have that potential.
  • 8:40 - 8:44
    Many illegal drugs have medical uses.
  • 8:44 - 8:47
    So for non-psychedelics
    such as MDMA, ecstasy,
  • 8:47 - 8:50
    where there is good evidence
    in post-traumatic stress disorder
  • 8:50 - 8:53
    and also some studies
    going on in addiction.
  • 8:53 - 8:55
    And of course, there is cannabis
  • 8:55 - 8:57
    where we have a range
    of different disorders
  • 8:57 - 9:01
    from pain, spasticity, cancer, epilepsy,
  • 9:01 - 9:04
    inflammatory diseases,
    and also sleep disorders.
  • 9:04 - 9:08
    All of these, potentially, are amenable
    to treatment with cannabis.
  • 9:10 - 9:12
    So, why don't we use these drugs?
  • 9:12 - 9:18
    That's because the WHL and the UN
    have said they are too dangerous,
  • 9:18 - 9:19
    which is certainly untrue.
  • 9:19 - 9:21
    I can tell you categorically
  • 9:21 - 9:24
    none of our patients died
    in the experiments we did on them.
  • 9:25 - 9:28
    Most of our governments
    perpetuate this lie.
  • 9:28 - 9:32
    And many of us - hopefully, not you -
    have closed minds.
  • 9:32 - 9:36
    We do not want to believe
    that these might have therapeutic utility.
  • 9:37 - 9:39
    So I want to say to you now,
  • 9:39 - 9:43
    surely now: you, if not everyone,
  • 9:44 - 9:45
    should accept the fact
  • 9:45 - 9:50
    that these drugs potentially
    could be very important medicines.
  • 9:50 - 9:54
    For the sake of the millions of people
    in the world who could be helped,
  • 9:54 - 9:56
    it's time to say there should be no limits
  • 9:56 - 10:01
    to the therapeutic research
    we do with these drugs.
